AbstractThe entry of nanocarriers into solid tumors poses an intrinsic size paradox in enhanced permeability and retention (EPR)-mediated passive delivery: larger nanoparticles accumulate more but face diffusion barriers, while smaller ones penetrate deeper but are rapidly cleared. Transcytosis-mediated active penetration resolves this trade-off by evading diffusion resistance, realizing simultaneous efficient accumulation and deep penetration.
